What is recommended; to run the pre-amp playing music at low or high volumes? will 100 hours be enough?

The amount of output voltage provided by the volume control doesn't matter to break in. The pre amp is running with its gain stages seeing constant voltages at all times and the volume control only acts as a valve to allow so much output voltage to be delivered to the pre amp output jacks.
